Brucine and Strychnine is a terpene indole alkaloid. Strychnine acts as an antagonist of glycine and acetylcholine receptors. It binds noncovalently to the same receptor, preventing the inhibitory effects of glycine on the postsynaptic neuron. Therefore, action potentials  are triggered with lower levels of excitatory neurotransmitters. When the inhibitory signals are prevented, the motor neurons are more easily activated

Brucine’s mechanism of action closely resembles that of strychnine. It acts as an antagonist at glycine receptors and paralyzes inhibitory neurons.

Glycine binds to receptors on inhibitory neurons to terminate action potentials. Its binding terminates an action potential by causing an influx of chloride ions into the neuron, repolarizing the neuron to its resting potential.

Nux vomica works by regulating various process of the body including action potential and is useful for treating debility, paralysis, dyspepsia, dysentery, chronic rheumatism, weakness of limbs, sexual weakness. It is therapeutically recognized for strengthening of spinal cord and nerves.

Pharmacological action of piperine has been proved to be shown due to its interaction with various transmitters, i.e., neuropeptides: substance P, neurokinins and calcitonin gene related peptide. 

Vanilloid compounds like piperine bind to specific receptor sites which results in a decreasing the levels of transmitters of pain sensation such as substance P, and certain amino acids like glutamate and aspartate
